Upgrade Advanced EPR/DEER Spectroscopy Facility

2019/20 • $150,000 • PI,CO
Electron paramagnetic resonance (EPR) and double electronelectron resonance (DEER) spectroscopy are important structural biology methods to obtain information on protein dynamics and protein conformations, which is complementary to cryo-electron microscopy (cryo-EM) and x-ray crystallography....

Chaperones and Proteases of the Plasmodium falciparum Parasite

2014/15-2018/19 • $235,000 • PI
Protein homeostasis in the cell is regulated by a wide array of quality control systems consisting of molecular chaperones and proteases. These systems assist newly-translated proteins in folding to their native state and also ensure that misfolded proteins and short-lived regulatory proteins...

Characterization of the structure and stability of human serum albumin isolated using a novel plasma extraction technique****

2018/19 • $25,000 • PI
Molecular chaperones are a highly interactive group of cellular proteins that fulfill central roles in all aspects of protein homeostasis including protein folding, assembly, and unfolding of substrates. Chaperones are typically categorized into families based on their sequence similarity and...

High resolution characterization of macromolecular structure and dynamics

2016/17 • $145,571 • PI,CO
Funds requested in this NSERC Research Tools and Instruments (Category 1) grant application will be used to purchase a state-of-the-art and new-to-market Size Exclusion Chromatography Multi-Angle Static Light Scattering (SEC-MALS) system and to upgrade a multiuser analytical ultracentrifugation...

An Enhanced Combinatorial Super-Resolution Microscopy Platform

2015/16 • $150,000 • PI,CO
RTI funding will be used to enhance the spatial, temporal, and spectral resolution of an exceptionally capable super-resolution combinatorial microscopy platform that combines the sub-nanometer 3-D resolution of scanning probe microscopy with super-resolution fluorescence microscopy. This latter...

Core facility infrastructure for optimizing conditions for lipidic cubic phase-based membrane protein crystallization

2014/15 • $150,000 • PI,CO
Membrane proteins perform a variety of biological functions that are critical to the survival of bacteria, plants, and eukaryotes. Crystal structures of these membrane proteins have shed enormous insights into the functions of transporters, signal transduction receptors, and enzymes. However,...

Chaperones and proteases of the Plasmodium falciparum parasite

2013/14 • $30,000 • PI
The eukaryotic protozoan parasite Plasmodium falciparum is the causative agent of malaria. It harbors two organelles of prokaryotic origin, the mitochondrion and the apicoplast. The apicoplast is a non-photosynthetic plastid derived from an ancient red algal endosymbiont. Hence, the apicoplast...

Systematic Analysis of the Regulation of Hsp90 by its cofactors

2012/13 • $26,000 • PI
Protein homeostasis in the cell is regulated by a wide array of molecular systems consisting of molecular chaperones and proteases. These systems ensure that proteins fold to their native state and maintain their proper conformation throughout their lifetime in the cell. The chaperone...

The present invention is directed to activators of cylindrical proteases ("ACPs"), particularly ClpP, and the role thereof in the diagnosis and treatment of bacterial infections. A number of ACPs were identified that activate caseinolytic protease P ("ClpP"), which independently can only degrade small peptides. In the presence of ACPs, ClpP may be activated to allow it to degrade larger proteins, hence, abolishing the specificity arising from the ATP-dependent chaperones. Members of the ACPs...
